The Amsterdam American Hotel offers 175 guestrooms, junior suites and four suites on six floors and the rooms are an important example of the authentic architectural Art Nouveau style with original stained-glass windows and elegant granite vanities. All 124 Classic American rooms, 36 Deluxe American rooms, 13 American Junior suites and four suites will be non-smoking rooms. Some of the rooms offer a Leidseplein view, canal view, or a balcony.
All guestrooms, junior suites and suites are equipped with air conditioning, cable and satellite TV, pay TV, radio, alarm clock, working desk with lamp, direct dial telephone with voicemail, dataport, high speed internet access, iron with ironing board, minibar, safe and art deco styled bathroom with bathtub, bathrobes, hairdryer and European toiletries.

The Classic American Rooms have a size between 20 to 30 squaremetres (215 to 323 square feet) and they are located throughout the hotel. These rooms are equipped either with one king size bed, one queen size bed or two twin size beds and many rooms offer stunning views over the Leidseplein or the famous canals of Amsterdam and some rooms have a balcony.

The Deluxe American Rooms have a size of 30 to 35 squaremetres (323 to 377 square feet) and they are located thoughout the hotel. These elegant rooms are luxuriously appointed and equipped either with one king size bed, one queen size bed or two twin size beds. Many of these rooms offer panoramic view of the Leidseplein or the famous canals of Amsterdam. Some of these larger guestrooms offer a separate sitting area or original stain-glass windows and can have a balcony. The bathrooms feature extra luxurious bathroom amenities.

The American Junior Suites have a size between 35 to 40 squaremetres (377 to 431 square feet) and they are located throughout the hotel. These elegant rooms are luxuriously decorated and equipped either with one king size bed or one queen size bed and offer a Lavazza Espresso machine. Many of the junior suites offer panoramic view of the Leidseplein or the famous canals of Amsterdam. These spacious rooms offer a separate sitting area and some offer original stain-glass windoews and can have a balcony. Some suites offer a fire place. The bathrooms feature extra luxurious bathroom amenities.

The Mata Hari Suite is named after the Dutch spy, courtesan and dancer, who lived from 1876 to 1917 and spent a part of her honeymoon in the Amsterdam American Hotel. This spacious and elegantly decorated suite has a size of 58 squaremetres (625 square feet) and features a large king size bed. The two corners with sofa and comfortable chairs are perfect for an intimate dinner or small informal meetings. The working desk is hidden from view. This suite offer a stereo system and guests can enjoy views of the Leidseplein or the Leidsecanal. The spacious and luxurious bathroom is equipped with a whirlpool bathtub.

The Amsterdam American hotel offers two Split-Level Suites. The new Ruud de Wild Art Suite is decorated in a modern style in the colours of white and black and this suite is named after after Ruud de Wild, a radio DJ and artist, who has designed this two-level suite. This luxurious suite offers one king size bed and a comfortable sitting area and a elegantly equipped bathroom.
